AstraZeneca’s Phase III DESTINY-Breast05 trial showed ENHERTU significantly improves invasive disease-free survival in HER2-positive early breast cancer patients at high risk of recurrence. ENHERTU, an engineered HER2-directed ADC, reduced the risk of disease recurrence or death by 53% compared to T-DM1, with 92.4% of patients alive and free of invasive disease at three years. It also improved other key secondary endpoints, including disease-free survival, distant recurrence, and brain metastases. Overall survival data were not mature at the interim analysis. The safety profile of ENHERTU was consistent with known data, with no new safety concerns identified.
