Allogene Therapeutics presented Phase 1 trial data for ALLO-316, showing a 31% response rate in patients with advanced or metastatic RCC. The CAR T product demonstrated expansion, persistence, and tumor infiltration while maintaining a manageable safety profile. Patients with CD70 TPS ≥50% saw a 31% confirmed response rate, with four of five responders remaining in response, including one in remission for over 12 months. The trial also showcased effective diagnostic and management strategies for mitigating IEC-HS. ALLO-316’s transformative promise in solid tumors offers hope to patients with limited treatment options.
The ongoing Phase 1 TRAVERSE trial evaluates ALLO-316, an AlloCAR T™ product targeting CD70, in patients with advanced or metastatic clear cell RCC. The FDA has granted ALLO-316 Regenerative Medicine Advanced Therapy (RMAT) designation for addressing unmet needs in RCC. With a goal of delivering readily available cell therapy on-demand, Allogene is developing a pipeline of “off-the-shelf” CAR T cell products for cancer and autoimmune disease. The company’s innovative Dagger® technology enables robust CAR T cell expansion and deep, durable remissions in solid tumors.
