Olema Pharmaceuticals announced promising results from a Phase 1b/2 study of palazestrant with ribociclib in ER+ HER2- advanced breast cancer. The combination showed a median PFS of 15.5 months across all patients in the 120 mg palazestrant cohort. Among patients with prior CDK4/6i treatment, median PFS was 9.2 months in ESR1 wild-type tumors and 13.8 months in ESR1 mutant tumors. The combination was well-tolerated with no new safety signals. Data supports the ongoing Phase 3 OPERA-02 trial for frontline advanced or metastatic breast cancer. These findings will be presented at the ESMO Congress 2025.
